So discovering the chemicals the body uses has been done with both animal and human tissue. The next step in studying these chemicals was to develop drugs that aided or interfered with their action. Beta-blockers, such as propranolol, interfere with receptors thus lowering heart rate and blood pressure and thus are used to treat hypertension. Betaagonists, such as isoproterenol, stimulate receptors increasing heart rate and blood pressure and as such are prescribed when a person is in heart failure. But while the basic chemical properties were usually the same across species lines, the problem arose when intact animals were then used to test the drugs developed based on the discoveries. The serotonin agonist sumatriptan is a prime example. Serotonin acts as a constrictor on blood vessels. It was thought that a drug derived from serotonin would aid in treating migraine headaches as the cause of migraines was thought to be blood vessel dilation in the head. Constrict the dilated blood vessels and the migraine goes away. Based on experiments with dog saphenous veins, researchers isolated a receptor they thought was responsible for blood vessel constriction in the head and only the head. If a person takes a drug that constricts the blood flow in vessels in the head, that is fine provided those vessels are dilated and provided no other vessels are constricted e.g., those in the heart. So sumatriptan was developed and tested extensively on animals. No severe side effects were reported. Humans however did have problems. Many people with no heart disease what so ever took the drug for their migraine and experienced chest pain because sumatriptan was not only constricting their blood vessels in their head but also the ones supplying oxygen to their hearts. Many experienced heart attacks and some died. Slight differences between species accounted for this effect. Has human tissue been used to test the drug, this problem might have been avoided. All the discoveries about serotonin could have been made using human tissue and when the drug was ready to be tested, had human tissue been used then also the species differences would not have occurred and the drug would have been modified to avoid coronary artery constriction. There was no reason to use animals, either intact or their tissue at anytime. In the long run, all it did was mislead researchers and cause human deaths. It is important to note however that the above examples and indeed essentially everything in this category, involve either research on isolated tissues or research on simple organisms like flies. The controversy in society today surrounding using animals in research usually revolves around the use of intact animals and usually such use is for drug testing or research into human diseases. When justifying the use of animals in research the vested interest groups tend to lump all uses into one category since they can justify using animal tissues and simple organisms like flies but cannot justify the more controversial use of intact animals as human models. We try very hard to separate the areas where animals are scientifically useful from the areas where they are not. This is the nature of genuine honest inquiry and science in general.

Fierce, continuous pain occurs in the orbit and face several times daily as brief attacks over periods of several weeks, separated by attack-free intervals of complete remission. The word attack is used to describe each onset of pain and the word episode or cluster for the period comprised of series painful attacks followed by remission. Diagnostic criteria include A : at least 5 attacks meeting criteria B and D; B : severe unilateral orbital, supraorbital and or temporal pain lasting 15 to 180 minutes without treatment; C : headache associated with at least one of the following manifestations occurring on the side pain of the conjunctival redness, tearing eyes, nasal congestion, rhinorrhea, sweating of the face and forehead, myosis, ptosis, edema of the eyelid; D : frequency of 1 to attacks per day; E : at least one of the following characteristics : medical history, physical and neurological examinations are not suggestive of any organic disorder; an organic disorder exists but the first cluster headache was temporally independent of its. Prevalence is unknown in France, but is estimated to be 1 10, 000 inhabitants based on foreign studies. The first attacks occur between ages 10 and 30 in 2 patients. An underlying genetic factor is probable. The pathophysiology of this disease remains unknown. At present, treatment is only symptomatic. The only two treatments of attacks with proven efficacy are subcutaneous sumatriptan and oxygen inhalation. Keywords Contunuous facial pain, vascular algesia of the face headache, attack: onset of pain and tadalafil.
Human ureter strips could be antagonized by ketanserin. In addition, combined administration of the 5-HT4 receptor antagonist DAU 6285 and the 5-HT3 receptor antagonist ondansetron caused a rightward shift of the cumulative concentration-response curve of 5-HT. Given individually, the 5-HT receptor antagonists methiothepin with higher anity to 5-HT7 than to 5-HT1A, ondansetron and DAU 6285 were unable to antagonise the contractions evoked by 5-HT. Message Emphasize the importance for older patients of keeping their healthcare professionals informed about their use of herbs. Barriers to revealing this information may include their embarrassment or fear that healthcare providers may: Discount their self-care efforts Not understand the importance of herb use in their culture Be insulted because they lack faith in the doctor's skill IV. Questions and Answers You may wish to break into a one-to-one or brown-bag format to review individual medications and provide counseling. If so, advertise ahead of time, so participants know to bring their medications and their herbal therapies, or at least a list. If you choose a traditional question-and-answer format, be sensitive to the personal nature of the questions--you may wish to answer some, but defer more personal questions to one-to-one discussions after the talk. Similarly, if one participant asks multiple or very involved questions about a case, arrange a one-to-one session later.
Special dietary instructions: do not increase your consumption of foods containing vitamin k, such as liver, green leafy vegetables, broccoli, and cauliflower, without discussing your diet with your doctor, for example, sumatriptan in pregnancy.

Starobrat-Hermelin B, Kozielec T. Department of Family Medicine, Pomeranian Medical Academy, Szczecin, Poland. Magnes Res 1997 Jun; 10 2 ; : 149-56 Children with ADHD are 'a group at risk' as far as their further emotional and social development and educational possibilities are concerned, and the consequences of the lack of an appropriate therapy appears to be serious. Some of these children do not respond to prevailing therapy methods. It is reported that dietetic factors can play a significant role in the etiology of ADHD syndrome, and magnesium deficiency can help in revealing hyperactivity in children. The aim of our work was to assess the influence of magnesium supplementation on hyperactivity in patients with ADHD. The examination comprised 50 hyperactive children, aged 7-12 years, who fulfilled DSM IV criteria for ADHD syndrome, with recognized deficiency of magnesium in the blood blood serum and red blood cells ; and in hair using atomic absorption spectroscopy. In the period of 6 months those examined regularly took magnesium preparations in a dose of about 200 mg day. 30 of those examined with ADHD showed coexisting disorders specific to developmental age, and 20 of them showed disruptive behaviour. The control group consisted of 25 children with ADHD and magnesium deficiency, who were treated in a standard way, without magnesium preparations. 15 members of this group showed coexisting disorders specific for developmental age, and 10 members showed disruptive behaviour. Hyperactivity was assessed with the aid of psychometric scales: the Conners Rating Scale for Parents and Teachers, Wender's Scale of Behavior and the Quotient of Development to Freedom from Distractibility. In the group of children given 6 months of magnesium supplementation, independently of other mental disorders coexisting with hyperactivity, an increase in magnesium contents in hair and a significant decrease of hyperactivity of those examined has been achieved, compared to their clinical state before supplementation and compared to the control group which had not been treated with magnesium. Is attention-deficit hyperactivity disorder an energy deficiency syndrome? Todd RD, Botteron KN. Department of Psychiatry, Washington University School of Medicine, St. Louis, Missouri 63110, USA. Biol Psychiatry 2001 Aug 1; 50 3 ; : 151-8 Attention-deficit hyperactivity disorder ADHD ; is a highly heritable yet clinically heterogeneous syndrome associated with hypocatecholamine function in subcortical and prefrontal cortical regions and clinical response to medications that enhance catecholamine function. The goal of this article is to present a hypothesis about the etiology of ADHD by synthesizing these findings with recent experiments indicating that activity-dependent neuronal energy consumption is regulated by cortical astrocytes. The scientific literature was searched from 1966 to the present using MEDLINE and relevant key words. Inattention and impulsivity may be related to hypofunctionality of catecholamine projection pathways to prefrontal cortical areas, resulting in decreased neuronal 295.
