Murine parvoviruses are common infectious agents in laboratory mice. Minute virus of mice MVM ; causes acute infection, whereas mouse parvovirus MPV ; can persist for extended periods. Therefore, it is important to differentiate natural infections caused by these agents. The primary detection method for parvovirus infection in laboratory mice is serology to detect serum antibodies to these agents. Many serological tests, including tests that detect antibodies to the nonstructural NS ; protein of the virus, determine that an animal has been infected but cannot identify the causative serogroup. Furthermore, NS-based tests require virus replication in host cells for levels of NS protein expression that elicit a detectable humoral response. In contrast, virus capsid VP ; proteins, which form the virus particle and convey serogroup specificity, can begin to elicit immune responses when the virus infects the host. We have developed an MPV ELISA that detects antibodies to the viral capsid protein VP2, and determined the test sensitivity and specificity using sera from experimentally and naturally infected mice. The VP2 protein from MPV strain 1a was cloned and expressed in E.coli, purified by His-tag chromatography, and used as antigen in an ELISA assay. Sera were tested at 1: 50 and 1: 100 dilutions by VP2 ELISA and at 1: 10 dilution by immunofluorescence assay IFA ; using MPV infected L3 cells. Of 504 sera screened, 91 were MPV positive by IFA and 87 of those also were positive by ELISA 4 false negatives ; . There was one false positive serum by ELISA. These results calculate to a test sensitivity of 95.6% and specificity of 99.8%. Of the 87 MPV positive sera, 35 were negative when tested against MVM VP2 antigen. For 23 of the 52 remaining sera, the OD450nm against MPV VP2 antigen was between 20% and 49% higher than against MVM antigen. For 28 sera, it was greater than 50% higher against MPV antigen than against MVM antigen. Further diagnostic testing, including comparative tests of sera using the MPV VP2 ELISA and NS ELISA, is being performed. The MPV VP2 ELISA offers a sensitive, serogroup specific test to detect and classify parvovirus infection in laboratory mice. The antigens used for capturing antibodies in the sera can be made conventionally by producing whole virus particles or by expressing recombinant proteins in prokaryotic or eukaryotic system. Recombinant Histagged antigens for parvovirus, arenavirus and coronavirus were cloned using the baculovirus expression system. Recombinant antigens were produced in insect cells and purified from cell lysate using nickelchelating chromatography procedure. Purified antigens were used for coating the 96-well ELISA plates. Field trials were run using rodent sera samples for parvovirus NS-1 ELISA. Out of total 3457 rodent samples tested for NS-1 antibodies only 88 2.5% ; were found to be positive and 9 0.3% ; gave tissue control TC ; non-specific results. Although NS-1 ELISA is highly sensitive and specific in detecting NS-1 antibodies, the same may not be present in all the seropositive and sumycin.

Adiponectin, but not c-reactive protein, is associated with serum hormone binding globulin in the polycystic ovary syndrome PCOS ; . Chen Y-D I, Trader B, Azziz R CRP C-reactive protein ; is an inflammatory marker secreted by the liver, shown to predict the incidence of coronary artery disease and diabetes. The levels of CRP are associated with obesity, and are often elevated in PCOS, although it is unclear whether this increase is independent of the degree of obesity. Adiponectin APM ; is insulinsensitizing anti-inflammatory adipocyte-derived hormone. Most reports have demonstrated decreased levels of APM in PCOS, although it is unclear whether this occurs independent of obesity. This study has as its aim to investigate whether the levels of CRP and APM in women with PCOS differ from controls, independent of obesity. Eight groups of women, as noted in the Tables below, were examined. Different age groups were included as a surrogate assessment of the duration of the clinical and sub-clinical conditions. Serum CRP and APM were determined by ELISA; DHEAS by RIA, and SHBG by a competitive binding functional assay. Data were analyzed by ANCOVA, adjusted for age and BMI followed by ANOVA post hoc pair comparisons in SAS. Multiple regression models were also employed to evaluate the predictors of levels of CRP and APM. Our results indicate that: 1 ; women with PCOS had higher CRP levels p 0.02 ; , but not significantly different APM p 0.22 ; , than those seen in controls, regardless of age and body mass index BMI 2 ; obese individuals had higher CRP p 0.001 ; and lower APM p 0.002 ; levels; 3 ; CRP levels, but not APM, were higher in older subjects p 0.005 and 4 ; regression models inclusive of age, BMI, groups, CRP APM, SHBG, and DHEAS indicated that age p 0.002 ; and BMI p 0.001 ; predicted CRP in this study population. Only SHBG p 0.02 ; predicted APM. We conclude that obese individuals with or without PCOS possessed high inflammatory risk high CRP and low APM levels ; , and the difference in CRP remained after adjustment for BMI and age. Furthermore, SHBG activity was positively associated with APM levels, suggesting that low SHBG levels may play a pivotal role in the IR of PCOS.
